Heparin

1, anticoagulant: heparin or bis-oxalate anticoagulant.
2, Wintobe (Wen's) than the volume of the tube, the inner diameter of this tube 3mm long about 10mm, scale to 1mm intervals and 10cm can be about 1ml of blood. 3, 5ml syringe.
4, capillary tube (its capillary section is only about 12cm) caliber is not greater than 2mm.
Second, the experimental operation.
1、Prepare anticoagulation vials, add 0.5ml of bicuculline solution or 0.5-1mg of heparin in each vial, and bake it dry for spare use. 2、Use a dry syringe to draw 2-3ml of blood, inject it into the above vials, and shake it gently.
3、Use capillary burette to inject the above shaken anticoagulated blood into the wintrobe tube, to the scale "0", the bottom of the capillary burette tube will be slowly dripped into the blood, with the rise of the blood plane, and gradually upward extraction of the capillary burette, so as to avoid the production of air bubbles, and the mouth of the tube tightly plugged with a small rubber stopper. 4、The blood will be perfused with wintrobe tube to a relative centrifugal force of 2260g, 3000 rpm for 30 minutes.
5. Remove the Wintrobe and read the height of the RBC column.
Calculation: RBC ratio (%) = (height of RBC column / height of whole blood column) × 100% reference value:
Adult male: 0.4-0.50
Adult female: 0.37-0.48
1-3 years 0.35-0.47
Newborn 0.4-0.67
