DNase Free Grade: Overview

DNase Free grade refers to biological reagents that have been verified to be free of deoxyribonuclease (DNase) enzymes. DNases degrade DNA by cleaving the phosphodiester bonds between nucleotides. Contamination with DNases can compromise experiments involving:

· RNA isolation or analysis (e.g., RT-PCR)

· DNA-free RNA preparation

· Cloning and sequencing

· Forensic or diagnostic workflows

To ensure the integrity of such processes, reagents, buffers, water, and consumables used must be DNase Free or even DNase/RNase-free.


Common DNase Free Reagents

· Ultra-pure water treated (e.g., by autoclaving and filtration) to remove DNases and often RNases.

· Used to resuspend RNA or sensitive enzymes.

· Often used in RNA isolation or nucleic acid handling.

3. DNase Free Enzymes

· Certified nuclease-free pipette tips, tubes, and filter tips.

· Avoids cross-contamination from laboratory consumables.


Certification and Quality Control

Manufacturers typically confirm DNase Free status using:

· Functional assays (e.g., incubating test DNA with the reagent and assessing degradation).

· ELISA-based or fluorometric detection for trace nucleases.

· Production in cleanroom environments or under ISO-certified conditions.


Applications
