Descripción general

  Orcein, also known as lichenin, has a CAS number of 1400-62-0 and a molecular weight of 500.51. It is a biological mordant that can be used for staining HBsAg, flagella, and other structures.

  Lichen Red Ethanol Solution (1%), also known as Acidified Orcein Staining Solution, is mainly prepared from orcein, hydrochloric acid, ethanol, deionized water and other components. The solution is acidic, featuring simple operation and high specificity, while the staining effect may vary with different batches of raw materials. It can be used for the staining of Hepatitis B Virus Surface Antigen (HBsAg), as well as the staining of lipofuscin and elastic fibers. This reagent is intended for research use only and not for clinical diagnosis or any other purposes.

Operating Procedures (For Reference Only):

1. Fix the tissues in 10% neutral formalin fixative, followed by routine dehydration and embedding.

2. Cut the sections at a thickness of 4 µm, then perform routine dewaxing to water with xylene or environment-friendly dewaxing and clearing solution.

3. Immerse the sections in acidified potassium permanganate solution (self-prepared) for oxidation for 5 minutes, then rinse briefly with water.

4. Transfer the sections into 1% oxalic acid solution (self-prepared) for bleaching for 2 minutes to remove the coloration caused by potassium permanganate, followed by rinsing under running water for 2 minutes.

5. Rinse the sections briefly in 70% ethanol.

6. Place the sections into orcein ethanol solution, cover them, and stain for 5 hours or longer.

7. Rinse the sections twice in 70% ethanol, 10–30 seconds each time, until no staining solution elutes from the sections.

8. Conduct routine dehydration, clear the sections with xylene or environment-friendly dewaxing and clearing solution, and mount the sections with neutral gum.

1. The staining effect may vary depending on different batches of raw materials.

3. Please use the reagent as soon as possible after opening to avoid affecting the results of subsequent experiments.
