Descripción general

Lignin is a biopolymer with a three-dimensional network structure formed by the interconnection of three phenylpropane units via ether and carbon-carbon bonds. It is present in lignified tissues and primarily functions to harden cell walls by forming an interwoven network, serving as a major component of the secondary wall. Lignin is mainly located between cellulose fibers, providing compressive strength. In woody plants, lignin constitutes about 25% of the biomass and is the second most abundant organic compound in the world. In nature, lignin is often linked with cellulose and hemicellulose, forming a Lignin-Carbohydrate Complex (LCC). Consequently, it is currently not possible to isolate native lignin with its structure completely intact.


Principle

The Lignin Staining Solution (Methyl Red Method) utilizes methyl red to stain lignin. After staining, lignified cell walls appear yellow. This staining method is a simple microchemical technique for detecting lignin in plant cell walls. However, the stain fades over time and is therefore not suitable for preparing permanent slides. This reagent is for research use only, not for clinical diagnosis or other purposes.

Procedure (For Reference Only)

1. Place the prepared sectioned material on a glass slide. Add 50-100 µL of Methyl Red Staining Solution and allow it to sit briefly to permeate the material.

2. Place a cover slip on top and observe under a microscope.


Staining Result

Lignified cell walls appear yellow.


Precautions

1. The greater the degree of lignification, the deeper the color.

2. It is generally recommended to use free-hand sections or frozen sections to preserve the integrity of the sample contents.

3. The use of paraffin sections should be avoided as much as possible, as the complex paraffin embedding process may lead to lignin loss.

4. The stain fades over time and is therefore not suitable for preparing permanent slides.

5. After opening the reagent, please use it as soon as possible to prevent affecting subsequent experimental results.
