Low-density lipoprotein is one of the serum proteins, primarily synthesized by the liver. It is positively correlated with the occurrence of coronary heart disease and the development of atherosclerotic lesions, serving as a key indicator for the classification of lipid disorders and the prediction of associated risks.
Testing Principle: A precipitating agent is used to isolate low-density lipoprotein cholesterol from serum. Cholesterol ester is hydrolyzed into free cholesterol and free fatty acids under the catalysis of esterase, thereby converting cholesterol ester into FC. Subsequently, cholesterol oxidase catalyzes the oxidation of FC to produce △4-cholestenone and H₂O₂. Finally, peroxidase catalyzes the oxidation of 4-aminoantipyrine and phenol by H₂O₂ to generate red quinone compounds, which exhibit a characteristic absorption peak at 500 nm.
