LumiDye™ Dibenzocyclooctyne-streptavidin (DBCO-SA) is a ready-to-use click chemistry conjugation reagent, prepared by covalently modifying high-purity streptavidin with DBCO-PEG5-NHS ester. This product enables rapid and efficient copper-free strain-promoted alkyne-azide cycloaddition (SPAAC) with terminal azide-modified antibodies, oligonucleotides, polymers, or other molecules under mild conditions.
Streptavidin possesses distinct biological properties: each molecule consists of four subunits capable of binding four biotin molecules with extremely high affinity, exhibiting a binding constant as high as 10¹⁵ M⁻¹, making it widely applicable in areas such as biological detection, separation, and targeted delivery, and many other fields. DBCO (dibenzocyclooctyne) is a cyclooctyne that undergoes efficient, copper-free click chemistry with azides under mild conditions. This reaction features high selectivity, fast kinetics, and excellent biocompatibility, making it suitable for live-cell and in vivo applications. The introduction of a polyethylene glycol (PEG) spacer enhances the reagent’s solubility, stability, and biocompatibility, reduces non-specific binding, and prolongs circulation half-life. DBCO-SA combines the advantages of streptavidin and DBCO groups. In biomolecular labeling and conjugation workflows, DBCO-SA first utilizes the specific binding between streptavidin and biotinylated molecules, followed by the click reaction between the DBCO group and azide-modified molecules, enabling accurate linkage of various biomolecules. This provides a convenient tool for biological research, and the entire process occurs under mild conditions with minimal impact on the native activity of the biomolecules.
The DBCO-SA was purified by size chromatography to remove non-conjugated molecules and ensure adequate applications both in-vitro and in-vivo.

Usage Suggestion
Storage conditions: The lyophilized powder should be stored dry and protected from light at 2–8°C for long-term preservation. After reconstitution, the solution is recommended to be stored at 4°C for no more than 1 week, or aliquoted and frozen at –20°C.
