Hemoglobin binds to oxygen to form oxyhemoglobin, which is converted to deoxyhemoglobin upon deoxygenation. By measuring the absorbances of oxyhemoglobin, deoxyhemoglobin, and methemoglobin at 560 nm, 576 nm, and 630 nm, respectively, and applying the extinction coefficients of each component at these specific wavelengths, the percentage mass concentration of methemoglobin can be calculated. Combined with the total hemoglobin content determined from the sample, the absolute methemoglobin content can then be calculated.
