Most of these methods utilize the principle that Alcian Blue, a cationic dye, can bind to acidic groups-specifically, Alcian Blue forms insoluble complexes with anionic groups (e.g., carboxyl groups and sulfate groups) present in tissues.
The Mucin Staining Solution (Azure A Metachromatic Method) leverages the metachromatic property of Azure A. After staining, the color of the stained tissue or tissue components differs significantly from the original color of the dye complex, creating a distinct contrast. The post-staining treatment of sections is crucial for the stability of metachromatic staining, and ethanol is recommended for preservation. This reagent is for research use only and not intended for clinical diagnosis or other purposes.
Materials to Be Prepared by the User
1. A series of ethanol solutions, distilled water, deionized water, xylene or environment-friendly dewaxing and clearing solution, and mounting medium

Operating Procedures (for Reference Only)
1. Dewax sections to water using xylene or dewaxing solution (Cat. No. D292661).
2. Stain with Mucin Staining Solution (Azure A Metachromatic Method) for 10 minutes, then rinse with deionized water for 5 minutes.
3. Dehydrate with gradient ethanol solutions, clear with xylene or dewaxing solution (Cat. No. D292661), and mount sections with mixed mounting medium.
Staining Results
Acidic mucins and proteoglycans: Red to purple
