N-arachidonoyl phosphatidylethanolamine (NAPE) is a precursor in anandamide (AEA) biosynthesis via multiple pathways (1). NAPE can be cleaved by a phospholipase D yielding phosphatidic acid and AEA (1,2). On the other hand, in macrophages NAPE is cleaved by a phospholipase C yielding phosphoanandamide (pAEA). A phosphatase, PTPN22 is one enzyme that cleaves pAEA leading to the generation of anandamide. Another pathway proceeds via double deacylation of NAPE generating glycerophospho-NAE. Phosphodiesterase-mediated cleavage of this intermediate yields anandamide. N-Arachidonoyl phosphatidylethanolamineis a precursor in anandamide biosynthesis
