La N-hexadecanoil-L-homoserina lactona es una de las AHL lipofílicas de cadena lateral larga producidas por el homólogo de la AHL sintasa LuxI, SinI, implicado en la señalización de quórum en S. meliloti, una bacteria fijadora de nitrógeno. Es la AHL más abundante producida por la proteobacteria R. capsulatus y activa el intercambio genético entre células de R. capsulatus. La N-hexadecanoil-L-homoserina lactona tiende a localizarse en entornos celulares relativamente lipofílicos de las bacterias y no puede difundirse libremente a través de la membrana celular. Las lactonas de N-acilhomoserina de cadena larga pueden ser exportadas de las células por bombas de eflujo o pueden ser transportadas entre células comunicantes por vesículas extracelulares de membrana externa.
