The Papanicolaou method is commonly used for routine cytological staining. Papanicolaou Stain was initially used only to assess estrogen levels in vaginal epithelium and detect pathogens such as Candida and Trichomonas in the genital tract. At present, most laboratories use ready-to-use staining solutions; therefore, the optimal modified conditions for each staining solution should be noted. The final cytoplasmic staining should be transparent and visible, and nuclear chromatin should be easily distinguishable. Currently, modified Papanicolaou staining solutions contain various ions and exhibit polychromatic staining effects. After staining, the cytoplasm is bright and highly transparent, with clear structures of the nuclear membrane, nucleolus, and chromatin.
Orange G6 Staining Solution is one of the components of Papanicolaou Stain, mainly composed of Orange G and phosphotungstic acid. It is often used in combination with EA36 or EA50 to stain the cytoplasm bright green, blue, and pink. It is frequently used for staining gynecological cytological smears, such as screening for cervical cancer and precancerous lesions, and is also suitable for staining non-gynecological cytological samples including pleural effusion, ascites, and sputum.
Procedure (For Reference Only):
1. Perform the procedure according to the specific experimental requirements.
2. Stain with Orange G6 Staining Solution for approximately 2 minutes.
Precautions:
1. All staining solutions require filtration and should be replaced regularly.
2. For your safety and health, please wear a lab coat and disposable gloves during operation.
