Phenylalanine hydroxylase (PAH), also known as phenylalanine 4-hydroxylase (P4H) and phenylalanine 4-monooxygenase (Phe 4-monooxygenase), with an EC number of EC 1.14.16.1, belongs to the family of aromatic amino acid hydroxylases.PAH is a stable enzyme that plays a key and rate-limiting role in phenylalanine metabolism and requires tetrahydrobiopterin as a cofactor.With tetrahydrobiopterin as the cofactor, phenylalanine hydroxylase catalyzes the hydroxylation of phenylalanine, converting phenylalanine to tyrosine and producing dihydrobiopterin.It can also convert tryptophan to 5-hydroxytryptophan (5-HTP) in the presence of the tetrahydrobiopterin cofactor.
