Descripción general

Plant tissue culture technology, also known as plant aseptic culture technology or in‑vitro culture technology, is based on the theory of plant cell totipotency. Under suitable artificial culture medium and environmental conditions, the isolated plant organs (such as roots, stems, leaves, flowers, shoot‑tips, fruits, etc.), tissues (such as cambium, epidermis, surface layer, pith cells, endosperm, etc.), cells (megaspores, microspores, somatic cells, etc.) and protoplasts can be induced to produce callus, adventitious buds and adventitious roots, and finally develop into intact plants. Prior to medium preparation, to facilitate operation, simplify procedures, ensure accurate dosage and reduce the time‑cost and weighing error of chemical components, inorganic macro‑elements, micro‑elements, iron salts, organic substances and hormones required for culture media are usually prepared separately as concentrated stock solutions with several‑fold higher concentrations. Corresponding volumes of each stock solution can be pipetted as pre‑calculated during medium preparation.

Plant Nutrient Solution‑Trace Element Stock Solution (400×) is mainly composed of potassium iodide, boric acid, manganese sulfate, zinc sulfate, copper sulfate, sodium molybdate, etc. It can be used to prepare common plant nutrient solutions such as Hoagland’s solution for plant tissue culture and plant growth‑rate detection. Dilute to 1× before use and supplement with other components. 100 mL of this stock solution can be used to prepare 40 L of nutrient solution. This reagent is intended for research‑only applications and is not suitable for clinical diagnosis or other purposes.

Materials to Be Prepared:

1. Distilled water or deionized water, dilute acid, dilute alkali, macro‑element stock solution, iron‑salt stock solution, organic substances, hormones.

2. pH meter, magnetic stirrer, beaker.

Operating Procedure (For Reference Only):

1. Take a 1‑L beaker, add 800 mL deionized water, then pipette 2.5 mL Micronutrient Stock Solution (400×). While stirring, add macro‑element stock solution, iron‑salt stock solution, organic substances, hormones and other components sequentially. Adjust the pH to 5.5‑5.9 with dilute acid or dilute alkali, then top‑up the volume to 1 L to obtain the target nutrient solution. The prepared nutrient solution is valid for 1 month when stored at 4 ℃.

2. If half‑strength (1/2 dosage) is required, halve the dosage of this reagent.

Precautions:

1. Select a suitable culture medium according to different materials and plant species. Optimal conditions are preferably determined experimentally.

2. The recommended pH range of nutrient solution is 5.0‑6.5. For strict sterility, perform autoclaving at 121 ℃ for 15‑20 min.

3. Use up the reagent promptly after opening to avoid adverse effects on subsequent experiments.
