Descripción general

  Poly-L-lysine Solution, abbreviated as PLL, refers to Poly-L-lysine hydrobromide in its chemical form. Its molecular formula is L-Lys-(L-Lys)n-L-Lys•xHBr, with a molecular weight ranging from 150,000 to 300,000, and its CAS Number is 25988-63-0.

  PLL is an adhesive commonly used for coating glass slides. It can be directly diluted and applied to experiments related to cell or tissue culture. Poly-L-lysine with a molecular weight greater than 70,000 can promote the adherent growth of cells. This reagent is suitable for enhancing cell adhesion and nucleic acid hybridization. The prepared slides can be stored at 4℃ for six months. This reagent is for research use only and is not intended for clinical diagnosis or any other purposes.

Operating Procedures (For Reference Only)

1. For Cell Culture

① Dilute the Poly-L-lysine Solution to an appropriate concentration according to experimental requirements before use. The coating time, concentration of Poly-L-lysine Solution, and even the selection of diluent may vary for different cell types. Please refer to relevant literature to perform coating appropriately.

② When using Poly-L-lysine Solution for cell culture, the coating duration should be at least 5 minutes. Some experiments require coating for 1-2 hours, while others may need overnight coating.

③ After coating, aspirate the Poly-L-lysine Solution and air-dry the culture vessel until completely dry as observed by the naked eye. Drying can be accomplished by blowing air in a fume hood for several minutes; certain experiments may require drying for 2 hours or longer. Generally, an extended drying time is more conducive to subsequent cell adhesion.

④ Proceed with cell culture. Alternatively, the coated vessel can be rinsed with an appropriate solution such as water, PBS, or cell culture medium prior to cell seeding.

2. For Nucleic Acid Hybridization

① Method 1: Take the pre-prepared glass slides or coverslips, cool them to room temperature after heating at 160℃, dip them into and out of the Poly-L-lysine Solution several times, air-dry them, and store at 4℃ for later use. They can also be stored at room temperature for up to one month.

② Method 2: Apply the Poly-L-lysine Solution onto the slides and air-dry; the coated slides are ready for use in cell smears and tissue sections.

③ Method 3: Add 5-10 μl of Poly-L-lysine Solution onto a slide, then use another coverslip to spread the solution using the blood smear technique, or press another slide tightly against it and rub them against each other to coat the facing surfaces with the solution.

Precautions

1. Poly-L-lysine can be digested and absorbed by certain cell types. Excessive intake of Poly-L-lysine may induce cytotoxicity.

2. Avoid repeated freezing and thawing. For large-volume usage, dispense an appropriate amount of the solution and store it at 4℃, with a validity period of 6 months.

3. When dipping slides into the Poly-L-lysine Solution, ensure that the slides are fully submerged in the liquid; otherwise, incomplete coating may cause sample detachment.

4. Prevent dust contamination during the drying process.

5. For your safety and health, wear a lab coat and disposable gloves during operation.

6. Please use the reagent as soon as possible after opening to avoid affecting the results of subsequent experiments.
