Descripción general

  Ponceau S is a commonly used protein stain. As a negatively charged reagent, it binds to both positively charged amino acid residues and non-polar regions of proteins, producing distinct red bands on blotting membranes. This binding is reversible, allowing the stain to be washed off with pure water or PBS buffer after use without interfering with subsequent experiments. This product is designed for detecting proteins on PVDF or nitrocellulose (NC) membranes in Western Blot experiments to assess transfer efficiency. It offers high sensitivity and ease of use, with a detection limit of 250 ng for proteins.

Protocol

  1. After protein transfer, fully immerse the PVDF or NC membrane in Ponceau S staining solution. Agitate on a shaker for 3–5 minutes.

  2. Rinse the membrane 2–3 times with pure water, PBS buffer, or another appropriate solution (3–5 minutes per rinse) to remove unbound dye until clear protein bands appear.

  3. After detection, rinse the membrane again 2–3 times with pure water, PBS buffer, or another suitable solution (3–5 minutes per rinse) to remove Ponceau S bound to proteins.

  4. Proceed with standard Western Blot detection steps.

Precautions

  1. Ponceau S staining solution is not suitable for nylon membranes.

  2. Wear a lab coat and disposable gloves for safety.
