Pyruvate carboxylase (PC, EC 6.4.1.1) is widely present in the mitochondria of animals, molds, and yeasts. It catalyzes the conversion of pyruvate, ATP, CO₂, and water to oxaloacetate, ADP, and inorganic phosphate (Pi). As the first rate-limiting enzyme of gluconeogenesis, it plays a crucial role in maintaining the dynamic balance of blood glucose.
Assay Principle: PC catalyzes the formation of oxaloacetate, ADP, and Pi from pyruvate, ATP, CO₂, and water. Malate dehydrogenase further catalyzes the conversion of oxaloacetate and NADH to malate and NAD⁺. The activity of PC is reflected by measuring the oxidation rate of NADH at 340 nm.
