Regula el cierre del tubo neural cefálico, inhibe el crecimiento de neuritas y la ramificación de neuronas corticales, así como la formación de sinapsis maduras. La unión a su receptor NEO1/neogenina induce la activación de la vía de señalización RHOA-ROCK1/Rho-quinasa a través de la cascada UNC5B-ARHGEF12/LARG-PTK2/FAK1, lo que provoca el colapso del cono de crecimiento neuronal y la inhibición del crecimiento de las neuritas. Además, la unión de RGMA a NEO1/neogenina conduce a la inactivación de HRAS al influir en la vía HRAS1-PTK2/FAK1-AKT1.
