Descripción general

Sudan dye is more soluble in lipids than in solvents. When the dye contacts lipid-containing samples, a large amount of dye penetrates into lipid structures and stains them red.


Instructions for Use

1. Rinse coverslips 3 times with Hanks solution.

2. Fix samples with formaldehyde-calcium fixative for 20 minutes.

3. Rinse coverslips 3 times with distilled water.

4. Wash coverslips 3 times with 95% ethanol.

5. Fully cover specimens with Sudan III staining solution and stain for 10-30 minutes.

6. Rinse coverslips 3 times with 95% ethanol.

7. Mount slides with glycerol gelatin and observe.

Precautions:

1. Prepare formaldehyde-calcium fixative personally: mix 10 mL formaldehyde and 1 g calcium chloride, add water to make 100 mL solution.

2. Conduct preliminary tests with 1 to 2 samples before formal use.

3. Wear lab coat and gloves for personal protection during operation.
