Tris-Acetate-EDTA Buffer (TAE) is the most commonly used buffer for DNA electrophoresis in laboratories.
The standard composition of 1× TAE working solution is: 40 mM Tris-Acetate, 1 mM EDTA (pH 8.0).
TAE Buffer (50×, Powder) is primarily composed of 2 M Tris-Acetate and 50 mM EDTA. Before use, it should be dissolved in distilled or deionized water to prepare a 50× concentrated stock solution. This stock solution is then diluted to 1× for use as the working solution.
This reagent is for research use only. Not for use in diagnostic procedures or other purposes.
Procedure (For Reference Only)
1. Preparation of 50× TAE Concentrated Stock Solution
Take approximately 700 mL of distilled or deionized water.
Add Reagent A and Reagent B sequentially, stirring thoroughly until completely dissolved.
Bring the final volume up to 1000 mL with water. This yields the 50× TAE Buffer stock solution.
2. Preparation of Working Solution
Dilute the prepared 50× TAE Buffer stock solution to 1× for use in DNA electrophoresis experiments.
Notes
1. Reagent A is provided as a powder. Reagent B is liquid at room temperature but may solidify at lower temperatures; it can be dissolved by warming in a water bath if needed.
2. If the single-use volume is small, it is recommended to aliquot the reagent after preparation to reduce contamination risk from repeated opening.
3. Please wear laboratory appropriate attire and disposable gloves during operation to ensure experimental safety.
4. Use the reagent as soon as possible after opening and keep it sealed to avoid affecting subsequent experimental results.
