The cations in Toluidine Blue O exert the staining effect, and acidic substances in tissue cells combine with these cations to be stained. Toluidine Blue O also contains two auxochromes that promote the ionization of the dye to form salts, enhance the staining power of chromophores on tissues, and thus stain the tissue cells on sections-specifically, it can stain cell nuclei blue. Clinically, Toluidine Blue O is frequently used to stain chondrocytes and mast cells.
Toluidine Blue O Stain Solution (For Cells) is suitable for staining acidic mucopolysaccharides in cells, especially for staining sulfur-containing acidic mucous substances, which appear red after staining.
Operating Procedures (For Reference Only):
1. After preparing the cell smear, add Toluidine Blue Staining Solution (Cell-Specific) dropwise and stain for 5 minutes.
2. Add an equal volume of distilled water to the smear, mix well, and stain for another 15 minutes.
3. Rinse with water, air-dry the smear, and examine under a microscope.
Staining Result:
Acidic mucopolysaccharides in cells appear red.
Precautions:
1. To verify the staining effect, blood smears from chronic myeloid leukemia (eosinophils) or bone marrow smears from aplastic anemia (tissue eosinophils) can be used as controls.
