UltraBio™ 2× Taq Platinum Universal SYBR Green Premix (ROX Separated) is a ready-to-use master mix optimized for quantitative PCR (qPCR) using SYBR Green I dye-based chemistry. It incorporates antibody-mediated hot-start AK Taq V2 Polymerase and a specially formulated buffer, ensuring high specificity, excellent amplification efficiency, and superior detection sensitivity.
This product includes separate vials of ROX reference dye, making it compatible with a wide range of real-time PCR instruments. Users only need to add primers and template to perform the reaction.
Intended Use: Quantitative or qualitative detection of nucleic acids using DNA templates. Simply add primers and RNase-free water to complete the reaction.

Protocol
1. Reaction Setup
1.1 Thaw and mix all PCR reagents thoroughly. Briefly centrifuge to bring the solution to the bottom of the tube. It is recommended to aliquot the master mix to avoid repeated freeze-thaw cycles.
1.2 Prepare the PCR reaction according to the table below:

Note 1: The amount of primers and template can be adjusted according to experimental needs.
Note 2: 50× ROX Reference Dye 1 is recommended for instruments requiring a high concentration of ROX, such as ABI PRISM® 7000/7300/7900HT, 7900HT Fast, StepOne, and StepOnePlus. 50× ROX Reference Dye 2 is recommended for instruments requiring a low concentration of ROX, such as ABI 7500 and 7500 Fast.
1.3 Mix thoroughly by pipetting gently or vortexing, as the solution is viscous. Briefly centrifuge to collect the liquid at the bottom of the tube.
1.4 Place the reaction tubes in a pre-programmed real-time PCR instrument and start the run.
