Descripción general

  The UltraBio™ Agarose Precast Gel (TAE) produced by Aladdin, also known as an agarose precast gel or nucleic acid precast gel, is a safe, convenient, and high-quality ready-to-use agarose precast gel commonly used for electrophoresis detection of DNA or RNA samples. The product offers stable quality, ease of use, and significantly improves experimental efficiency.

Instructions for Use

  1. Remove the UltraBio™ Agarose Precast Gel (TAE) from the packaging bag and place it, along with the tray, into the electrophoresis tank. Assemble the electrophoresis setup.

  2. The wells side corresponds to the negative electrode. Add 1× TAE buffer to the tank until the liquid level just covers the gel surface. Remove any bubbles from the wells if present.

  3. Add Loading Buffer to the DNA sample, mix thoroughly, and slowly pipette the sample mixture into the gel wells. Avoid piercing the bottom of the wells during loading. Use the same method to load the Marker. The recommended loading volume is 20–30 μL.

  4. Connect the power supply: red for the positive electrode, black for the negative electrode. DNA samples migrate from the negative to the positive electrode (the end with the wells is negative). Set the voltage to 120–250 V. Electrophoresis time varies with the size of the tank; larger tanks require longer run times.

  5. Run the gel under constant voltage (e.g., 150 V) until the bands reach the desired positions.

  6. After electrophoresis, turn off the power supply, remove the gel (with the tray), and place it directly under a gel imaging system to observe and photograph the bands. The non-toxic dye pre-incorporated in the gel has spectral properties similar to those of EB and can be observed and imaged under UV or fluorescence (594 nm).

  7. Clean, dry, and store the electrophoresis equipment after use.

Notes

  1. This product is pre-loaded with the latest non-toxic nucleic acid dye. No additional dye is required in the buffer. After electrophoresis, the gel can be directly imaged. The dye is less sensitive to single-stranded DNA or RNA compared to double-stranded DNA.

  2. Imaging exposure parameters:

    • Mode: Reflection

    • Light source: UV

    • Exposure time: 1000 ms

  3. To remove the gel from the tray, insert a knife or any flat tool under the gel and lift it from the U-shaped tray.

Precautions

  1. Using freshly prepared buffer for electrophoresis can significantly improve results. Note that repeated use of electrophoresis buffer may reduce ionic strength, increase pH, and decrease buffering capacity, potentially causing blurred bands and irregular DNA migration.

  2. Do not store below 0°C. Gels may freeze, leading to bubbles and cracks, rendering them unusable.

  3. This product is intended for scientific research by professionals only and must not be used for clinical diagnosis or other inappropriate purposes. Do not store in residential areas.

  4. For your safety and health, wear appropriate personal protective equipment and lab attire during operation.
