Bilirubin is a type of bile pigment and the main pigment in human bile. It serves as a crucial clinical indicator for diagnosing jaundice and assessing liver function.
This kit employs the Harrison method (also known as the Harrison tube test). The detection principle involves adsorbing bilirubin in urine using barium chloride, followed by adding Fouchet's reagent, which oxidizes bilirubin to biliverdin, producing a green color change. This kit offers high sensitivity, up to 0.05 mg/dL or 0.9 µmol/L, and is primarily used for the qualitative detection of bilirubin in human or animal urine. This kit is for research use only and is not suitable for clinical diagnosis or other purposes.
Operating Steps (For Reference Only)
1. Place 4 mL of fresh urine in a test tube and add approximately 2 mL of Barium Chloride Solution. Mix well; a white precipitate will form.
2. Centrifuge at 3000 g for 5 minutes. Discard the supernatant and retain the precipitate.
3. Add a few drops (approximately 50 µL per drop) of Fouchet's Reagent to the precipitate. The appearance of a green or bluish-green color indicates a positive result.
Precautions
1. Urine must be fresh. Test immediately after collection to avoid degradation of bilirubin due to light exposure.
2. The presence of salicylates or aspirin in urine may cause false-positive results.
3. Avoid adding excessive Fouchet's Reagent, as this may produce a yellow color and lead to false-negative results.
4. Please use the reagent as soon as possible after opening to avoid affecting subsequent experimental results.
