Descripción general

When proteins are far from their isoelectric point, they tend to form insoluble precipitates. Sulfosalicylic acid is a biological alkaloid reagent. Under acidic conditions, its anions can combine with positively charged proteins to form insoluble protein salts, resulting in precipitation. This product is a qualitative detection kit with a sensitivity of 0.05-0.1 g/L, primarily used for detecting proteins in human or animal urine. This kit is intended for research use only and is not suitable for clinical diagnosis or other purposes

Experimental Procedure

  1. Transfer 3-5 ml of fresh urine into a small test tube.

  2. Add 1-2 drops of Sulfosalicylic Acid Solution, allowing it to form a layer/interface.

  3. Observe immediately. If turbidity appears, it indicates the presence of protein in the urine. The depth of turbidity corresponds to the approximate concentration.

  4. Result Interpretation

Result Interpretation
Phenomenon Description
Estimated Protein Concentration
Negative (-)
No turbidity, clear and transparent
< 0.05 g/L
Suspicious (±)
Slight turbidity, faintly visible
0.05 - 0.2 g/L
(+)
Distinct white turbidity, no granules
~ 0.3 g/L
(2+)
Thin, milky turbidity with appearing granules
~ 1 g/L
(3+)
Milky and opaque, with flocculent precipitates
~ 3 g/L
(4+)
Flocculent precipitates, coagulated into clumps
> 5 g/L

Precautions

  1. If the urine sample is turbid, it should be centrifuged or filtered first.

  2. Highly alkaline urine may cause false-positive results.

  3. The sensitivity of this method is 0.05-0.1 g/L.

  4. Highly alkaline urine may also lead to false-negative results. Add a few drops of the Acidification Solution and retest.

  5. False-positive results can be caused by organic contrast agents, large doses of penicillin, or high concentrations of uric acid/urates in the urine.

  6. High concentrations of urea or urates in the urine may cause false-positive results.

  7. Use reagents promptly after opening to prevent potential effects on subsequent experimental results.
