Instructions for Use:
1.only trace amounts of peptides are deposited in the tube during the lyophilization. Before opening the tube, we recommend centrifuging the tube at approximately 8,000-12,000×g for 10-30 seconds to collect the peptides to the bottom of the tube.2.For P9813-25mg, dissolve the peptide in TBS completely by pipetting or flicking the tube gently. Do not vortex to avoid the denaturation of the peptide.3.Competitive elution of V5-tagged proteins from immunoprecipitated samples: This is a non-denaturing method with high elution efficiency and the eluted sample does not contain the Anti-V5 Tag antibody.a.Preparation of the V5 Tag elution buffer: Based on the experimental needs, prepare an appropriate amount of elution buffer by diluting the V5 Tag Peptide stock solution to 150μg/ml with TBS.b.Add 100μl of V5 Tag elution buffer (150μg/ml) to each immunoprecipitated sample and incubate on ice or at 4℃ with shaking for 0.5-2h. To increase the elution efficiency, the incubation time can be extended or the elution can be repeated. The volume of V5 Tag elution buffer is generally 5 times of the initial volume of the gel suspension or magnetic bead suspension.c.Centrifuge at 6,000×g for 30 seconds at 4℃ and carefully transfer the supernatant to a new tube. The supernatant contains the V5-tagged proteins or their protein complexes.4.Store the eluted V5-tagged proteins or protein complexes at 4℃ for immediate use, or -20℃ for long term storage.
