개요

20(S)-Hydroxycholesterol (20α-Hydroxycholesterol) is an allosteric activator of Smoothened (Smo) that activates Hedgehog (Hh) signaling. In an Hh-responsive reporter assay using NIH 3T3 cells, 20(S)-Hydroxycholesterol showed an EC50 of approximately 3 μM. Its effects on Smo are stereoselective, and it induces Smo accumulation in primary cilia. 20(S)-Hydroxycholesterol directly binds to the extracellular cysteine-rich domain (CRD) of Smo and activates downstream Gli signaling; it did not activate a Wnt pathway reporter gene in the corresponding reporter assay. 20(S)-Hydroxycholesterol also exhibits pro-osteogenic activity and, in combination with simvastatin, enhances Raf/MEK/ERK signaling, osteogenic differentiation, and bone regeneration.
