aTAG 4531 (CFT-4531) is a potent, selective, and onmechanism tool degrader of MTH1 with DC 50 value of 0.28 nM and K i value of 1.8 nM. The degradation activity is due to the intricate formation of the ternary complex between the MTH1aTAG, CRBN E3 ligase , and aTAG tool degrader. Red: MTH1aTAG inhibitor , Blue: CRBN ligand , Black: linker.
