Under the catalysis of β-hydroxybutyrate dehydrogenase, β-hydroxybutyrate is converted into acetoacetate. At the same time, oxidized coenzyme I (NAD⁺) is reduced to reduced coenzyme I (NADH). The β-hydroxybutyrate content in the sample can be calculated by measuring the increase in NADH at 340 nm.
