Principle

Entamoeba coli (Entamoeba coli) encapsulated iron hematoxylin-stained specimens: Entamoeba coli is a non-pathogenic amoeba, often together with lysed amoeba, so it is necessary to distinguish. The method of finding the capsule is the same as before, and then change the oil microscope observation. Its capsule is larger than that of the lysed tissue amoeba, and there are 1-8 nuclei in the capsule, and the one with 8 nuclei is the mature capsule. The characteristics of the nucleus are: the nuclear membrane is thicker, the inner edge of the membrane has a layer of uneven size, irregularly arranged chromatin grains, the nucleolus is large and parochial. Chromatoid bundles with sharp ends. Glycogen vesicles are vacuolated.

1. Tissue lysed intra-tissue amoebic trophozoite iron hematoxylin stained specimens.

Oil microscopic observation:It is a parasitic form of the disease in the intestinal wall of the human intestines or in tissues such as the liver and lungs. It is large in size. The diameter is about 20-40μm.

The cytoplasm is blue-gray, the ectoplasm is transparent, and the pseudopods are finger-like or tongue-like after staining with iron hematoxylin. The endoplasm is granular, and the erythrocytes are stained as

The erythrocytes were stained blue-black and varied in size. The structure of the nucleus was the same as that of its capsule.

2. Iron hematoxylin-stained specimen of amebic trophozoites from enteric luminal tissue.

Oil microscopic observation: It is a parasitic co-generated cystic type in the lumen of human colon. The diameter was 7-20 μm (average 13 μm). The inner and outer stroma were not clearly differentiated, and the inner stroma was full of food vesicles containing bacteria but not erythrocytes. The structure of the nucleus is the same as that of the large trophozoites.

3. Charcot-Leyden crystals (Charcot-Leyden crystal)

Acute amoebic crippling disease in the feces often have colorless transparent (dark blue after Su dye) rhombic crystals appear, in the diagnosis of some significance, found such crystals, should be carefully looking for amoebic trophozoites.

4. Colonic amoebic trophozoites iron hematoxylin stained specimens.

It is a non-pathogenic ameba.

Observed under oil microscope: it is larger (20-50 μm) than the ameba in the lysate. The inner and outer stroma are not clearly differentiated, the inner stroma is full of food vesicles, contains bacteria but not erythrocytes, and the nucleus has the same characteristics as that of its capsule.

5. Live amoebic trophozoites

From the fresh feces of amebic dysentery patients of pus and blood part of the material, immediately for saline smear, cover slips with high power microscope observation. Or, a small amount of culture was aspirated from an artificial culture maintained at 37°C and examined on a slide.
