Principle
The Mycoplasma pneumoniae Rapid Test Card detects Mycoplasma pneumoniae lgM antibodies in serum. The patient's serum sample is added to each of the two test wells, and when the sample diffuses along the membrane, 3 drops of anti-human lgM alkaline phosphatase complex, 3 drops of wash solution, and 2 drops of substrate are added, and the results are observed after 5 minutes. The quality control wells are quality control and immobilized with human lgM and the test wells are immobilized with Mycoplasma pneumoniae antigen. Positive reaction results in blue color and negative results in colorless.
Materials and Instruments
Patient Serum Move 1. Remove the kit from the refrigerator and equilibrate at room temperature for 20 minutes. 2. Remove the test card from the tinfoil bag and label it. 3, Use a pipette to add 2 drops of serum to the two test holes at the lower end of the card. 4. Incubate the card at 22-25°C for 2 minutes, within which time the serum should diffuse along the membrane to the upper test wells. 5, add 3 drops of enzyme conjugate to the lower two test wells, incubate at 22-25℃ for 2 minutes, the enzyme conjugate should diffuse along the membrane to the upper test wells. 6, add 3 drops of wash solution to the upper two test wells and wait for complete absorption. 7, add 2 drops of substrate reagent to the upper two test wells, incubate at 22-25℃ for 5 minutes, observe the reaction result immediately after 5 minutes. Caveat 1. Serum samples can be stored at 2-8°C for 3 days. If testing cannot be done during this period, store at -20°C. Samples should not be repeatedly frozen or thawed. 2. Label well during testing to prevent confusion. Common Problems Judgment of results: Judgment is based on the reaction results of the two reaction holes at the upper end: Positive result: blue color appears in both wells at the upper end. Suggests the presence of Mycoplasma pneumoniae lgM. Negative result: the quality control wells appear blue or slightly blue, and the test wells are colorless or dark gray, suggesting the absence of Mycoplasma pneumoniae lgM or its concentration is below the lower limit of detection. Invalid response: the quality control wells do not appear blue. Possible reasons for invalid reaction are: quality of reagents or test card, improper handling, poor diffusion of sample, severe contamination of enzyme conjugate, excessive lipid content in serum. Non-contaminated serum can be centrifuged and tested again . The kit does not cross-react with adenovirus, rheumatoid factor, respiratory syncytial virus, antinuclear antibodies, influenza A and B, parainfluenza 3, Histoplasma capsulatum, cytomegalovirus.
