Principle

Heating can denature proteins and solidify them, and the addition of acid can bring proteins close to the isoelectric point and cause them to precipitate. In addition, the addition of acid can also dissolve alkaline salts precipitation.

Materials and Instruments

Experimental reagents:

①, dilute acetic acid solution: take 5 ml of glacial acetic acid and add water to 100 ml

Experimental method:

1, take about 10ml fresh clear urine in a heat-resistant large test tube.

2. Place the test tube obliquely over a flame and boil the urine in the upper part.

3, add 3-4 drops of acetic acid, and then boil, boil immediately after the inspection results. If turbidity or precipitation, suggesting that the urine contains Pr.

Experimental results to determine:

Negative (a): not turbid

Trace (Shi): Slightly turbid 0.l-0.15g/L

Weakly positive (+): Clear white haze 0.2-0.5g/L

Positive (++): Clear turbidity with obvious particles 0.6-2.0g/L

Strong positive (+ + + +): a lot of flocculent precipitation turbidity 2.0-5.0g / L

Strongest positive (++++): clots with large amounts of flocculent precipitate >5.0gL

Caveat

1, the test is less interfering factors, detected sensitivity 0.15g / L, white, ball sensitivity is consistent.

2, add too much acid, causing a false-negative positive weakening.

3、No salt and low salt diet patients, false negative.
