Product Introduction

Malachite Green Phosphate Assay Kit is a rapid, convenient and highly sensitive kit for determining soluble free inorganic phosphate in solutions. It can be used for the quantification of phosphate concentration in various solutions.

The detection principle is based on the reaction that malachite green, molybate and free inorganic orthophosphate form a green molybdophosphoric acid complex. The absorbance of this green complex at the wavelength of 620–640 nm is directly proportional to the concentration of free inorganic phosphate in the solution.

This kit provides a reliable method for the high-sensitivity quantitative detection of free inorganic phosphate. However, it is only suitable for the direct determination of free phosphate in solutions. For phosphate bound to lipids or proteins, the phosphate must be released by appropriate methods prior to the assay.

Product Components

Operating Instructions

1.Preparation of Phosphate Standard Solution

Dilute the 5 mM phosphate standard solution to prepare a 50 μM phosphate working solution. For example, take 10 μl of the 5 mM phosphate standard solution and add 990 μl of standard diluent to obtain 1 ml of 50 μM phosphate working solution. Deionized water or double-distilled water is preferred as the standard diluent. The diluted working solution can be stable for at least one day at 4 ºC. If self-prepared reagents are used to dilute the standard solution, ensure that the reagents are free of phosphate contamination.

2.Preparation of Chromogenic Reagent

Calculate the required volume of chromogenic reagent based on the number of samples. The ratio of Malachite Green Reagent A to Malachite Green Reagent B for use is 36:17. The chromogenic reagent can be stored at 4 ºC in the dark and remains stable for one day.

Setup of Standard Curve

3.Prepare phosphate standard solutions at concentrations of 0, 1, 2.5, 5, 10, 20, 30, 40 and 50 μM according to the table below.

4. Determination of Phosphate Concentration

a. Add an appropriate volume of the sample to be tested into a 1.5 ml centrifuge tube. If the sample volume is less than 200 μL, make up the volume to 200 μL with standard diluent and record the dilution factor of the sample at this time.

b. Pipette 200 μL of phosphate standard solutions with different concentrations and samples to be tested from each tube in sequence into the sample wells of a 96-well plate.

c. Add 50 μL of chromogenic reagent to each well, mix thoroughly using a multi-well plate shaker or gently by pipetting up and down, and incubate at 37 ºC for 30 minutes.

d. Measure the absorbance at 630 nm using a microplate reader.

e. Calculate the phosphate concentration of the sample based on the standard curve and the dilution factor of the sample to be tested.

Notes

1.The chromogenic reagent must be prepared and used immediately. It can be stored at 4 ºC for one day after preparation. The reagent must be equilibrated to room temperature before use.

2.The detection wavelength of this experiment ranges from 620 to 660 nm, with 630 nm being optimal. If a microplate reader is not available, a spectrophotometer can be used instead. In this case, adjust the volumes of the chromogenic reagent, samples and standard solutions proportionally to ensure the final volume to be tested meets the minimum detection volume of the cuvette.

3.The sample volumes described in this instruction manual are based on a 96-well plate. If cuvettes or 384-well plates are used, the volumes should be adjusted proportionally.

4.Laboratory detergents contain high concentrations of phosphate. Ensure that the experimental utensils are thoroughly cleaned to prevent phosphate contamination.

5.For better detection results, it is recommended to set up duplicate or triplicate wells for all phosphate standard solutions and samples to be tested.

6.If the measured absorbance is low, it may be due to the low phosphate content in the sample. Consider increasing the sample dosage within the allowable range of this kit.

7.Malachite Green Reagent A is strongly acidic and corrosive. Handle it with care during operation, ensure effective protection to avoid direct contact with the human body, and take care to prevent corrosion of other items.

8.This product is intended for scientific research use by professionals only. It is not for clinical diagnosis or treatment, nor for use in food or pharmaceuticals, and should not be stored in ordinary residential premises.

9.For your safety and health, wear a lab coat and disposable gloves during operation.

Application Protocols

The following is a general microplate protocol for malachite green phosphate detection (literature guidance). For item-specific steps, refer to the product insert/CoA.

  • Materials: Malachite green phosphate reagent(s), inorganic phosphate standard (e.g., KH2PO4), phosphate-free water, compatible buffer (e.g., HEPES or Tris), clear 96-well plate.
  • Prepare Pi standards: Make a 1 mM Pi stock. Prepare 0, 2, 5, 10, 20, 30, 40, 50 µM standards in the same buffer as samples.
  • Samples: Quench enzymatic reactions to stop Pi generation (acid or heat) and cool to RT. Clarify by brief spin if turbid.
  • Assay setup (example volumes): Add 20 µL standard or sample to each well. Add 180 µL of color reagent. Mix gently by tapping or brief orbital shaking.
  • Incubate: 15–20 minutes at RT protected from light.
  • Read: Measure A620–650 nm. If dual-wavelength correction is available, reference at 750–800 nm to reduce scatter (instrument dependent).
  • Calculate: Subtract blank (0 µM) from all readings. Fit a linear regression of A versus [Pi] over the validated range. Determine sample [Pi] from the line, adjusting for any dilutions.

Quality controls:

  • Include at least one matrix-matched spike recovery (e.g., +10 µM Pi) per sample type; acceptable recovery typically 80–120% (laboratory criterion).
  • Replicates: Perform triplicates for standards and samples to estimate precision (CV%).

Note: Volumes/times may be adjusted to match the kit’s instructions if provided.

Biological Roles

This section describes the biological context of the analyte detected by the kit—orthophosphate (Pi)—not the kit reagents themselves (literature, general knowledge).

  • Central metabolite: Pi is essential for energy metabolism (ATP/ADP/AMP interconversion), glycolysis (e.g., glyceraldehyde-3-phosphate dehydrogenase step), oxidative phosphorylation, and substrate-level phosphorylation.
  • Signaling and regulation: Cellular phosphate levels influence kinase/phosphatase pathways indirectly through ATP availability and phosphorylation state turnover.
  • Structural roles: Phosphate esters stabilize macromolecules (DNA/RNA backbones, phospholipids). Free Pi participates in buffering capacity within physiological ranges together with H2PO4−/HPO42− systems.
  • Enzymology readouts: Many enzymatic reactions release Pi (e.g., ATPases, certain ligases in pyrophosphate-coupled reactions after PPi hydrolysis, phosphatases). Sensitive Pi quantification enables kinetic parameter determination (kcat, Km) and inhibitor screening.
  • Homeostasis: Organisms regulate Pi uptake, storage (e.g., polyphosphate), and mobilization via transporters and enzymes; deviations in Pi availability modulate metabolism and gene expression.

Implications for assay design:

  • Because Pi is ubiquitous, background contamination from buffers, glassware, or dust can be significant; rigorous control experiments and blanks are critical.
  • Mg2+ is commonly required for ATPase activity and is usually tolerated by colorimetric Pi assays; nonetheless, verify ion and chelator effects in your specific matrix.
Buffer Applications

This product is not a buffer. However, buffer choice is critical for accurate phosphate detection. The following guidance is general (literature) and can improve performance:

  • Avoid phosphate: Do not use PBS or phosphate-based buffers; even trace Pi elevates background and compresses the dynamic range.
  • Recommended buffers: HEPES, MOPS, PIPES, or Tris (pH 6.5–8.0) are commonly compatible. Minimize amines with strong absorbance near 620–660 nm and validate if using specialized buffers.
  • Metal ions: Mg2+/Mn2+ typically tolerated at enzymatic levels (e.g., 1–10 mM), but very high concentrations can influence complex formation. Confirm via spike-recovery.
  • Chelators: EDTA/EGTA can chelate molybdate; keep as low as feasible and verify impact at your working concentration.
  • Detergents: Keep nonionic detergents ≤0.01–0.05% where possible; validate background and linearity. Avoid cationic surfactants that may interact with cationic malachite green.
  • Proteinaceous samples: Proteins can precipitate upon acid addition; consider pre-quenching enzymatic reactions, then clarify by brief centrifugation before color development to reduce turbidity.

Practical recipes (general example):

  • 50 mM HEPES, 100 mM NaCl, 2 mM MgCl2, pH 7.5, filtered (0.22 µm). Prepare a phosphate-free water blank and a matched buffer blank for standard curves.
Green Alternatives

While colorimetric malachite green assays are widely adopted for Pi detection, users seeking greener or less hazardous approaches may consider the following (literature, general comparison):

  • Fiske–Subbarow (ascorbate-reduced) phosphomolybdate assay: Avoids malachite green dye but still uses molybdate and strong acid. Sensitivity is generally lower; reagents can be prepared from commodity salts, reducing packaging waste.
  • Enzymatic fluorescent Pi assays (e.g., phosphate-binding protein reporters): Aqueous, often neutral pH, and can minimize strong acids; however, they may require specialized instrumentation and can involve recombinant proteins and proprietary fluorophores.
  • Magnesium green/MDCC-PBP coupled systems: Reusable biosensors in some formats; lower hazardous acid usage, but cold-chain and biogenic materials are needed.
  • Ion chromatography or capillary electrophoresis: Solvent-light methods with minimal hazardous dyes; instrument-intensive and less convenient for high-throughput plates.

Tradeoffs:

  • Malachite green kits excel in simplicity, cost, and sensitivity at room temperature with standard plate readers.
  • Greener options may reduce acidic/azo/triphenylmethane dye waste but can increase cost, complexity, or require fluorescence capability.

Sustainability tips with this kit:

  • Prepare only the volume needed of working reagent.
  • Use microplate formats to minimize reagent consumption.
  • Segregate acidic/molybdate waste for proper treatment and consider waste minimization plans per institutional EHS.
Pharmaceutical Uses

This product is not intended for clinical or therapeutic use. No pharmacopeial monograph applies to the kit as a whole.

Relevant non-clinical, research/manufacturing contexts (general):

  • Enzyme characterization and QC: Quantifying ATPase/phosphatase activity in enzyme preparations, stability studies, and formulation screening.
  • Process development: Monitoring ATP consumption or inorganic phosphate release in cell-free biocatalysis workflows and in-process controls during enzyme production.
  • High-throughput screening: Colorimetric end-point readouts for inhibitor/activator discovery against phosphatases, kinases (via coupled assays), and NTPases using standard plate readers.
  • Analytical support: Assessing phosphate contamination in laboratory water or buffers to qualify reagents prior to sensitive assays.

Regulatory note:

  • For research use only (Product Data). Users in regulated environments should perform internal method validation (specificity, linearity, accuracy, precision, range, robustness) and document lot-to-lot qualification per SOPs. No clinical diagnostic or therapeutic claims are made or implied.
Physical Properties

Item-specific physicochemical specifications (e.g., concentration, pH, absorptivity) are not provided in the Product Data.

  • Appearance: Not specified for this item; refer to CoA/Spec Sheet.
  • Density, MP/BP, refractive index, logP: Not applicable for a multi-reagent aqueous kit.
  • Storage: 2–8°C, protected from light (Product Data). Shipped on wet ice (Product Data).

Assay readout properties (literature, general for malachite green Pi assays):

  • Measurement mode: Colorimetric.
  • Wavelength: Peak absorbance commonly in the 620–660 nm range; many protocols read at 620–650 nm with a broad bandpass.
  • Linear range (typical, plate-based): Often ~0.2–30 µM Pi; extended up to ~50–100 µM with pathlength/volume adjustments. Exact performance is kit/protocol dependent.
  • Limit of detection: Low sub-micromolar to ~1 µM Pi under optimized conditions; dependent on background and matrix.
  • Time to stable color: Frequently 10–30 min at room temperature; prolonged incubation may increase background due to non-specific color development.

Solubility/compatibility (literature, general):

  • Assay reagents are aqueous. The color complex forms in strong acid; organic solvents (e.g., >5–10% v/v DMSO, MeOH) and detergents can perturb dye–complex interactions and increase background.
  • Phosphate-containing buffers directly contribute to background. Use non-phosphate buffers for samples and standards.
Quality and Grades
  • Grade: BioReagent, Sterile (Product Data)
    • BioReagent grade indicates reagents suitable for biochemical/biological assays, emphasizing low levels of common biochemical contaminants that could interfere with enzyme activity or readouts (general definition).
    • Sterile indicates the kit solutions/components have been sterilized or sterile-filtered to minimize bioburden; this is important to limit phosphate contamination and microbial growth during repeated handling (general implication). It does not imply suitability for clinical/therapeutic use.

Item-specific notes:

  • UV cutoffs, metal limits, residual peroxide, and water content are not applicable or not provided for this kit. Not specified for this item; refer to CoA/Spec Sheet.
  • Performance characteristics (sensitivity, linearity, interferences) are assay- and matrix-dependent. For validation in regulated environments, generate a matrix-matched standard curve and document accuracy, precision, LOD/LOQ, and robustness.

Best practices to preserve quality:

  • Use only non-phosphate, high-purity buffers and water (e.g., freshly prepared ultrapure water) to minimize background.
  • Dedicate low-phosphate plasticware/glassware; pre-rinse with assay buffer or water.
  • Minimize repeated opening/closing of reagent bottles; if permitted by the CoA/SDS, aliquot under clean conditions to reduce contamination risk.
  • Protect reagents from light at all times to maintain dye integrity.
Reaction and Applications

This kit quantifies inorganic phosphate (Pi) released or consumed in biochemical reactions via malachite green–molybdate complexation (literature, general principle). Applications include:

  • Enzymology: Monitoring ATPase/GTPase activity, protein/alkaline/acid phosphatases, phospholipases (e.g., PLC), nucleotidases, kinase coupled assays (ATP consumption), and polymerase-driven pyrophosphate hydrolysis workflows (with pyrophosphatase).
  • Metabolic assays: Measuring Pi in glycolytic or oxidative phosphorylation studies, or in reconstituted systems consuming ATP.
  • Lipid and polymer chemistry: Quantifying phosphate in phospholipid dephosphorylation or surface phosphate release.
  • Environmental/analytical: Low-level orthophosphate in biochemical extracts or lab water (validation recommended for non-biological matrices).

Assay chemistry (literature overview):

  • In acidic medium, orthophosphate reacts with ammonium molybdate to form phosphomolybdic acid, which associates with malachite green dye to yield an intense green complex measured at ~620–660 nm.
  • The signal is directly proportional to Pi concentration within a defined linear range; sensitivity is typically sub‑micromolar to low micromolar depending on pathlength and background.

Practical tips (general):

  • Construct a fresh standard curve with inorganic phosphate (e.g., KH2PO4) spanning the expected sample range; matrix-match standards if possible.
  • Quench enzymatic reactions before adding color reagent (e.g., by acid addition or heat) to stop further Pi generation.
  • Avoid phosphate in buffers and plasticware; pre-rinse consumables. Validate tolerance to Mg2+/Mn2+ and chelators if present.
Reaction Conditions

The following conditions reflect general, literature-based guidance for malachite green phosphate assays and are not item-specific specifications.

  • Temperature: Ambient (20–25°C). Maintain consistent temperature across wells/replicates for reproducibility.
  • Incubation time: 10–30 minutes after adding color reagent, or until absorbance stabilizes. Excessive incubation can increase background.
  • Wavelength: Measure at 620–660 nm (commonly 620–650 nm) using a microplate reader or spectrophotometer.
  • Pathlength/format: 96- or 384-well plates (clear, flat-bottom) for high-throughput; cuvettes for highest accuracy. Shorter pathlengths reduce apparent absorbance—use standard curves in the same format.
  • Volumes (typical): 10–50 µL sample + 100–200 µL color reagent per well (96-well). Adjust to maintain reagent excess.
  • Quenching: Stop enzyme reactions before color development by acid addition or heat; cool to RT prior to adding reagent to minimize variability.
  • Standard curve: Prepare fresh Pi standards (e.g., 0–50 µM from a 1–10 mM KH2PO4 stock) in the same buffer as samples. Include blanks.
  • Interference control: Validate tolerance to salts (NaCl, KCl, MgCl2), chelators (EDTA), and detergents by spike-recovery. Avoid phosphate buffers.
  • Data analysis: Subtract blank, fit linear regression over the validated range, and report Pi concentration accounting for any dilution during assay setup.

Always consult the product’s IFU/CoA for any item-specific instructions.

Safety and Handling

Authoritative safety information must be taken from the SDS for this product. The Product Data do not list specific hazards or GHS classifications for the kit reagents.

  • GHS classification, signal word, H-statements, pictograms: Not specified for this item; refer to SDS.
  • Storage: Store at 2–8°C, protected from light (Product Data). Shipments on wet ice (Product Data).

General laboratory safety considerations (literature, typical for malachite green phosphate reagents):

  • Many malachite green phosphate reagents include strong mineral acid (e.g., HCl) and molybdate salts. Handle with appropriate PPE: lab coat, safety glasses, acid-resistant gloves. Use secondary containment to mitigate spill risk.
  • Work in a fume hood when handling acidic reagents or preparing bulk working solutions; avoid inhalation of vapors/aerosols.
  • Incompatibilities: Bases and phosphate buffers neutralize/consume the assay; oxidizing/reducing agents and strong detergents may interfere with color development.
  • First aid (general): In case of skin/eye contact with acidic solutions, rinse with copious water for at least 15 minutes and seek medical attention. If ingested or inhaled, follow institutional procedures and consult the SDS.
  • Waste: Collect acidic, molybdate-containing waste in labeled containers for hazardous disposal per institutional and local regulations. Do not discharge to drain unless permitted.

Always defer to the product-specific SDS and institutional EHS guidance.

Solvent Selection

This product is an aqueous colorimetric assay kit, not a solvent. Solvent selection pertains to the sample/buffer matrix used with the kit.

Buffer/solvent compatibility (literature, general guidance):

  • Avoid phosphate-containing buffers (e.g., PBS, phosphate, PhosSTOP) that contribute background Pi. Use non-phosphate buffers such as HEPES, MOPS, PIPES, or Tris for preparing samples and standards.
  • Ionic strength: Moderate salt (e.g., 50–200 mM NaCl) is usually tolerated, but very high salt can affect color development and baseline; verify by spike recovery.
  • Organic cosolvents: Keep DMSO/MeOH/EtOH as low as feasible (commonly ≤1–5% v/v) to maintain complex stability and spectral characteristics.
  • Detergents: Nonionic detergents (e.g., Tween-20, Triton X‑100) can raise background and scatter; if required, keep ≤0.01–0.05% and validate. Avoid cationic detergents that may interact with the cationic dye.
  • Chelators: High EDTA/EGTA can bind metals and perturb the molybdate complex; minimize or validate at intended concentrations.

Comparison (general):

  • Malachite green assay vs Fiske–Subbarow (ascorbate-reduced phosphomolybdate): MG offers higher sensitivity and microplate compatibility but is more susceptible to surfactants and organic solvents.
  • Malachite green vs fluorescent Pi probes: Colorimetric MG requires only a standard plate reader; fluorescent assays may offer greater dynamic range but higher cost and sensitivity to photobleaching.
Storage and Reconstitution
  • Storage conditions: Store at 2–8°C, protected from light (Product Data). Keep containers tightly closed when not in use. Avoid prolonged exposure to ambient light.
  • Shipping: Shipped on wet ice (Product Data). Inspect upon receipt; bring to 2–8°C storage promptly.

Reconstitution/preparation:

  • Specific reconstitution instructions (e.g., concentrate-to-working reagent, acid additions) are not provided. Not specified for this item; refer to CoA/Spec Sheet.
  • General guidance (literature): If components require mixing prior to use, equilibrate to room temperature, gently invert to mix (avoid foaming), and prepare only the volume needed for the day. Allow microbubbles to dissipate before dispensing to plates.

Handling and stability (general best practice):

  • Protect all dye-containing solutions from light during use; cover plates/tubes with foil.
  • Avoid microbial contamination; if permitted, aliquot into sterile containers to minimize repeated opening. Do not freeze unless the CoA explicitly allows it.
  • Discard reagents that show precipitate formation, unexpected discoloration, or altered background behavior.
  • For long assays, maintain consistent temperature to reduce drift. Document lot number, preparation date, and any dilutions on lab records for traceability.
