Rivalta Reagent, also known as Rivalta test reagent, is used for the qualitative differentiation of serous cavity effusions (such as pleural effusion and ascites), i.e., to distinguish between exudates and transudates. Detection Principle: Serous mucoprotein in serous cavity effusions is an acid glycoprotein with an isoelectric point of approximately pH 3–5, which precipitates in dilute acetic acid solution. During the test, the Rivalta reagent (dilute acetic acid solution) is mixed with the puncture fluid. If a white cloudy precipitate forms, the result is positive, indicating that the effusion is an exudate (commonly associated with inflammation, infection, or malignancy). If no precipitate forms or the precipitate rapidly dissipates, the result is negative, indicating that the effusion is a transudate (usually caused by non-inflammatory factors).
