Product description
RTGreen is a DNA binding dye used for real-time quantitative PCR (qPCR). The many advantages of this dye make it far superior to EvaGreen and Sybr Green. In addition to having similar spectral characteristics, RTGreen has three main characteristics that distinguish it from SYBR Green I.
1. The inhibitory effect of RTGreen on PCR is much less than that of SYBR Green I. Therefore, qPCR experiments using RTGreen can use a fast PCR step. At the same time, RTGreen can use a higher concentration in the experiment, so as to obtain a much stronger amplification signal than SYBR Green I. The higher concentration of RTGreen also eliminates the defect of "dye redistribution", making RTGreen not only used for multiplex PCR, but also for high resolution (high definition) melting curve analysis (HRM). This analysis is increasingly being used for genotyping and heteroduplex analysis after PCR. Due to the inhibitory properties of SYBR Green I on PCR, the use concentration must be very low. Therefore, SYBR Green I cannot solve the problem of dye redistribution caused by low concentration, and it cannot be used for multiplex PCR or HRM. At the same time, the problem of dye redistribution may also affect the reliability of conventional melting curves, because DNA strands with low melting points may not be detected for this reason.
2. RTGreen is extremely stable. It will not be damaged during normal storage, operation and PCR. The dye in the buffer solution can be safely stored at room temperature or in the refrigerator, or it can be repeatedly frozen and thawed. In contrast, SYBR Green I is unstable and has a stronger inhibitory effect on PCR after degradation.
3. RTGreen reduces cell membrane permeability, so it is safer than SYBR Green I. Independent laboratory test results show that RTGreen is neither mutagenic nor cytotoxic. On the contrary, although SYBR Green I itself is very weakly mutagenic, it may inhibit the normal DNA repair mechanism in cells, making it mutagenic and enhancing.
