Application Protocols

Per non-negotiable rules, only protocols explicitly provided by the product data can be listed as tested. No tested applications, dilutions, or conditions are specified for this item.

Item-specific tested applications:

  • Not specified for this item; refer to CoA/Spec Sheet.

General guidance (for planning only; not a claim of validation for this lot):

  • WB, IF/ICC, IHC, IP, and ELISA are common antibody applications. Start with standard titration series (e.g., 1:100–1:2,000) and optimize using positive/negative controls for ARRDC1.

For authoritative, lot-specific protocols (recommended dilutions, incubation times, species tested, positive control tissues/cell lines), consult the CoA/Spec Sheet.

Biological Roles

Focus on the target protein ARRDC1 (literature overview; no clinical claims).

  • Protein: ARRDC1 (Arrestin domain-containing protein 1)
  • Family: Arrestin-domain containing adaptors (ARRDCs), related to alpha-arrestins.
  • Cellular localization: Predominantly cytoplasmic and associated with the plasma membrane; implicated in endosomal and microvesicle pathways (literature).
  • Molecular function: Adaptor/scaffold protein that recruits components of the ubiquitin/protein trafficking machinery to regulate membrane protein internalization and sorting (e.g., Nedd4 family E3 ligases interactions; literature).
  • Biological processes: Involved in clathrin-independent endocytosis, receptor downregulation, and extracellular vesicle (microvesicle) biogenesis by mediating budding from the plasma membrane (literature). Can influence cargo selection into vesicles.
  • Pathways and interactions: Interfaces with ESCRT-related processes and ubiquitin-dependent trafficking. Reported to modulate cell surface receptor abundance and to facilitate shedding of ARRDC1-mediated microvesicles (ARMMs) that can transport signaling cargo between cells (literature).
  • Expression: Broad expression with tissue-dependent levels; regulation can occur via transcriptional and post-translational mechanisms (literature).

Research implications:

  • Antibodies to ARRDC1 enable mapping of protein abundance, localization (IF/IHC), and interaction networks (co-IP), and support studies on extracellular vesicle content. Validate species reactivity and application compatibility of this specific antibody lot via the CoA/Spec Sheet.
Buffer Applications

Antibody performance depends on appropriate buffer composition tailored to the assay. The following are general, literature-based guidelines; verify compatibility with your exact protocol and this Carrier Free antibody.

  • Western blot (WB):

    • Transfer/wash buffer: Tris–glycine or CAPS with 0.05–0.1% Tween-20.
    • Blocking: 5% nonfat dry milk or 3–5% BSA in TBS-T/PBS-T; BSA preferred if milk interferes with phospho-epitopes.
    • Primary antibody diluent: Same as blocking buffer but with reduced blocker (e.g., 1–3% BSA) to balance background vs. binding.
  • Immunofluorescence (IF)/Immunocytochemistry (ICC):

    • Permeabilization: 0.05–0.3% Triton X-100 or saponin (epitope-dependent).
    • Blocking: 1–5% BSA or serum from secondary host species in PBS/0.05% Tween-20.
    • Mounting media: Aqueous antifade media for fluorescence retention.
  • Immunohistochemistry (IHC):

    • Antigen retrieval: Citrate buffer pH 6.0 or Tris–EDTA pH 9.0 (heat-mediated), optimize per tissue/epitope.
    • Blocking/dilution: Similar to IF; include endogenous peroxidase quench if using HRP detection.
  • Immunoprecipitation (IP):

    • Lysis buffers: Non-denaturing (e.g., 1% NP-40 or Triton X-100 in TBS) preserve interactions; RIPA for stringent conditions. Include protease/phosphatase inhibitors.
  • ELISA:

    • Coating: Carbonate–bicarbonate buffer pH 9.6 or PBS; blocking with BSA/casein.

Item-specific recommended buffers, pH, and dilutions are not specified; consult the CoA/Spec Sheet.

Green Alternatives

While green chemistry primarily targets small-molecule processes, sustainable practices also apply to antibody use.

Greener practice options (general guidance):

  • Buffer selection: Prefer phosphate- or HEPES-based aqueous buffers over organic solvent systems. Avoid chlorinated solvents entirely.
  • Preservatives: Use minimal effective concentrations of preservatives (e.g., sodium azide) and consider sterile filtration/aliquoting to reduce bioburden rather than relying solely on chemical biocides.
  • Waste minimization: Aliquot upon receipt to extend shelf life and reduce disposals from degradation. Use refillable, low-adsorption plasticware where possible.
  • Energy savings: Store working aliquots at −20°C as recommended and avoid unnecessary deep-freeze storage when not required. Consolidate freezer space to reduce energy footprint.
  • Conjugation chemistry: When labeling, consider aqueous, copper-free click chemistries to reduce metal contamination and post-reaction cleanups.

Comparison (general):

  • Carrier-containing formulations vs Carrier Free
    • Carrier Free (this item): Lower background, easier conjugation, reduced protein-waste stream from carriers; requires more careful handling to prevent adsorption and aggregation.
    • With carriers (BSA/gelatin): Improved stability for routine handling but can introduce background and complicate MS or conjugation workflows.

Item-specific environmental certifications or solvent details are not specified; refer to institutional sustainability guidelines for implementation.

Pharmaceutical Uses

This product is provided strictly for research use only and is not intended for human or veterinary diagnostic or therapeutic applications.

  • GHS classification: Not specified for this item; refer to SDS.
  • Signal word: Not specified for this item; refer to SDS.
  • H-statements/Pictograms: Not specified for this item; refer to SDS.

General safety guidance (literature/best practice):

  • PPE: Wear lab coat, gloves (nitrile), and safety glasses when handling. Avoid ingestion, inhalation of aerosols, or skin/eye contact.
  • Biological origin: Antibodies are proteins typically produced in cell culture or animal hosts; handle as a biological reagent of low risk. Follow institutional biosafety procedures.
  • Asepsis: To maintain product integrity, work on ice where appropriate and use sterile, low-protein-binding plastics. Avoid microbial contamination.
  • Incompatibilities: Avoid extreme pH (<5 or >9), strong oxidizers, organic solvents, and surfactant concentrations that can denature proteins. Do not mix with proteases.
  • Freeze–thaw risk: Repeated freeze–thaw can cause aggregation and activity loss; follow Storage & Reconstitution guidance to aliquot upon receipt.
  • Spills/first aid: For small spills, absorb with inert material and clean with detergent. For eye/skin contact, rinse with water for ≥15 min; seek medical attention as needed. If inhaled, move to fresh air. If ingested, rinse mouth and seek medical advice. Always defer to SDS.

Waste disposal:

  • Dispose of unused material and contaminated consumables as laboratory bio/chemical waste per institutional and local regulations.
Solvent Selection

Antibodies are aqueous biological macromolecules; selection focuses on buffer systems rather than organic solvents.

General buffer selection (literature/practice):

  • Working buffers: PBS (10–50 mM phosphate, 150 mM NaCl, pH 7.2–7.6) or TBS (Tris 20–50 mM, 150 mM NaCl, pH 7.4–8.0) are commonly used to maintain isotonicity and stability.
  • Additives (if compatible with your assay):
    • Detergents: 0.05–0.1% Tween-20 for wash buffers to reduce nonspecific binding (avoid in conjugation steps).
    • Blocking proteins: 1–5% BSA, casein, or fish gelatin for blocking/dilution in immunoassays. Note: This product is Carrier Free; add blockers only in working buffers, not to the stock if you plan to perform conjugations.
    • Stabilizers: 5–10% glycerol or 0.1–0.5% trehalose can enhance freeze–thaw resistance (verify assay compatibility).
  • Preservatives: 0.02–0.05% sodium azide inhibits microbial growth (avoid with HRP or live-cell applications). Not included by default in Carrier Free products unless specified.

When to choose alternatives:

  • For metal-sensitive downstream assays (e.g., IMAC-MS), use metal-free buffers and low-salt formulations.
  • For live-cell work or functional assays, avoid azide and harsh detergents; use sterile, endotoxin-minimized buffers.

Item-specific solvent details (pH, excipients) are not specified for this product; refer to CoA/Spec Sheet.
