Application Protocols

Item-specific tested applications and recommended dilutions are not provided in the Product Data. Refer to the CoA/Spec Sheet for authoritative guidance.

General starting points (literature/common practice; not validated for this item):

  • Western blot (WB): Start at 1:1,000 for chemiluminescent detection; adjust 1:500–1:5,000 based on signal/background. Load lysate with verified ARRDC3 expression as positive control; include a knockout/siRNA control where possible.
  • Immunofluorescence (IF/ICC): Begin at 1:200 in blocking buffer; optimize fixation (e.g., 4% PFA) and permeabilization (e.g., 0.1–0.3% Triton X-100) as needed.
  • Immunohistochemistry (IHC-P): Trial 1:200 after antigen retrieval (e.g., citrate pH 6.0 or Tris-EDTA pH 9.0); fine-tune retrieval and blocking conditions empirically.
  • ELISA: Coating 0.5–2 µg/mL for capture; detection 0.1–1 µg/mL depending on substrate and instrument sensitivity.
  • Immunoprecipitation (IP): 1–5 µg antibody per 500–1,000 µg lysate protein; optimize lysis buffer stringency to preserve ARRDC3–partner interactions.

Controls and validation:

  • Include isotype controls and secondary-only controls as appropriate.
  • Verify specificity using genetic controls (knockout/knockdown) or overexpression with epitope-tagged ARRDC3.

These are general guidelines; tailor to your system and consult the product CoA/Spec Sheet when available.

Biological Roles

Focus: ARRDC3 (Arrestin domain-containing protein 3), the antigen recognized by this antibody.

General biology (literature, not item-specific):

  • Family: ARRDC3 is a member of the arrestin domain-containing (ARRDC) adaptor proteins related to β-arrestins.
  • Cellular role: Functions as an adaptor in membrane protein trafficking, often linking cargo receptors to endocytic and ubiquitination machinery.
  • Processes: Implicated in regulation of GPCR internalization and downregulation, interaction with E3 ubiquitin ligases, and coordination of endosomal sorting. May influence cellular signaling dynamics through receptor turnover and trafficking pathways.
  • Localization: Predominantly cytoplasmic with membrane association depending on cargo/receptor engagement; can localize to endosomes (literature context).
  • Experimental utility: Antibodies to ARRDC3 enable detection of protein expression, subcellular localization, and analysis of pathway modulation in cell and molecular biology studies.

Note: The above reflects general scientific literature and is provided for research context only. It does not constitute functional validation for this specific antibody lot. Consult the CoA/Spec Sheet for experimental validation data for SKU Ab089693.

Buffer Applications

Not applicable. This product is an antibody reagent, not a buffering agent or buffer system.

Practical note:

  • Prepare and use standard immunoassay buffers (e.g., PBS or TBS with appropriate blocking agents and detergents) according to your application. Refer to Application Protocols and your institutional SOPs.
Green Alternatives

Not directly applicable. Antibody reagents are aqueous protein solutions rather than organic solvents or reagents commonly evaluated for green chemistry substitution.

Sustainability considerations (general laboratory practice):

  • Minimize single-use plastics by planning aliquots efficiently.
  • Use phosphate-free or Tris-based buffers responsibly to reduce environmental phosphate loading when appropriate.
  • Consolidate cold-chain shipments where feasible; this product ships on ice packs per Product Data.
Pharmaceutical Uses

Not applicable. This product is designated for research use only and is not intended for human or veterinary therapeutic or diagnostic applications.

Formulation note (general for laboratory use):

  • Carrier-free antibodies are suited for custom conjugation and assay development. Any downstream use in GMP or clinical workflows would require separate qualification and is outside the scope of this product.

  • Solubility: Readily soluble in aqueous biological buffers (e.g., PBS) at mg/mL concentrations; avoid extreme pH or denaturants unless intended.
  • Stability: Improved by low temperature storage, neutral pH buffers, and inclusion of stabilizers (e.g., glycerol, sugars, proteins) where appropriate to the application; this item is listed as carrier free, so no stabilizing proteins are included by default.
  • UV absorbance: A280 ≈ 1.0 corresponds to ~0.7 mg/mL for IgG (literature approximation; extinction coefficient is sequence-dependent).

Note: For authoritative, lot-specific properties (concentration, buffer composition, extinction coefficient), consult the CoA/Spec Sheet.

Quality & Grades

This product is supplied as: Carrier Free, ExactAb™, Validated (per Product Data).

  • Carrier Free:

    • Meaning: Formulated without protein carriers such as BSA, gelatin, or other stabilizing proteins.
    • Implications: Reduces background in sensitive assays (e.g., mass spectrometry, conjugation chemistries, functional assays where carrier proteins interfere). May be more sensitive to adsorption or freeze–thaw—aliquoting is recommended.
  • ExactAb™:

    • Meaning (general): Denotes a recombinant or precisely defined antibody format produced under controlled conditions for sequence and lot-to-lot consistency.
    • Implications: Enhanced reproducibility across experiments; compatible with site-specific conjugation workflows depending on format. Consult CoA/Spec Sheet for clone/sequence/isotype details.
  • Validated:

    • Meaning: Internally qualified for defined research applications. Specific tested applications, species reactivity, and validation data are not listed in the Product Data; refer to the CoA/Spec Sheet for details.

What is NOT specified for this item (consult CoA/Spec Sheet):

  • Clone ID, isotype/subclass, host species, recombinant expression system, concentration, buffer composition, and any residual preservative content.

Practical notes:

  • For conjugation (e.g., HRP, fluorophores), carrier-free format is advantageous. Verify buffer components prior to conjugation to avoid reactive amines or azides if incompatible with your chemistry.
Reaction & Applications

This section (synthetic reaction chemistry) is not typically applicable to antibody reagents.

Relevant applications for this product involve immunoassays and protein detection rather than small-molecule transformations. See:

  • Target Specificity: antigen information for ARRDC3.
