Enterokinase (light chain, EK) is a recombinantly expressed serine protease that specifically recognizes the sequence Asp-Asp-Asp-Asp-Lys and cleaves the peptide bond at the C‑terminus of Lys. It can cleave fusion proteins containing this recognition sequence. It is used for research on recombinant fusion proteins or peptides in biopharmaceuticals, synthetic biology, and molecular biology. This product is a high-purity, high-specificity, high-activity mutant of bovine enterokinase recombinantly expressed and prepared in Escherichia coli. Physical form: Lyophilized powder containing 0.39 mg Tris‑HCl and 0.01 mg CaCl₂ Molecular weight: 26.2 kDa Reconstitution: Reconstitute with 50 μL ultrapure water to a concentration of 1 μg/μL pH range: Enterokinase shows optimal activity at pH 6.0–8.5 Digestion method: Recommended digestion buffer: 50 mM Tris or HEPES buffer, pH 6.0–8.5 For fusion protein samples: use a protease-to-protein ratio of 1:500 to 1:1000, adjust protein concentration to 0.2–0.5 μg/μL, and incubate at 4 °C–25 °C for 12 hours Notes: Digestion efficiency may be affected under the following conditions: 2 M urea, > 250 mM NaCl, > 20 mM β‑ME, > 0.1% SDS, > 50 mM imidazole, or PBS.
