Inoculation ring, inoculation needle preparation experiment
Inoculation ring, inoculation needle preparation experiment
The inoculation ring (needle), also known as the platinum ear (needle), is a common tool used in bacteriologic testing. Its correct use is an important basic technology. This experiment is derived from the laboratory instruction of Mudanjiang Medical College undergraduate 5-year testing program.

Principle
The inoculation ring (needle), also known as the platinum ear (needle), is a common tool used in bacteriologic testing. Its proper use is an important basic technique. Move The inoculation ring (needle) consists of three parts. Ring (needle) part of the platinum wire made of good, because easy to heat transfer and heat dissipation and not rust and durable. However, because of the high price, usually replaced by nickel wire. Commonly used ring diameter of about 2-4mm, needle length of 50-80mm, one end of which is fixed to the metal handle, the other end of the metal handle for the kink handle. Standard inoculation ring is in the ring filled with wet bacterial moss (Escherichia coli) weight of exactly 2mg, used to prepare a certain concentration of bacterial suspension or quantitative inoculation. Quantitative inoculation ring is to make the volume of the ring 1/100, 1/200, 1/300......1/1000 ml. It is used for quantitative culture of liquid.
The inoculation ring (needle) must be flame sterilized before and after use, usually by passing over the flame of an alcohol lamp or gas lamp, and can be used only after sterilization.
Flame Lamp:
Flame lamps are essential equipment for sterilizing inoculators during bacteriological test operations and for disinfecting test tube ports and other items. Gas lamp is more ideal inoculation sterilization equipment, the size of the flame can be adjusted according to the needs of their own. The flame can be divided into two layers: outer flame and inner flame. The outer flame is very hot due to rich contact with oxygen, so the outer flame should be used when cauterizing inoculation rings or articles. Where there is no gas source, alcohol lamps can be used, but the temperature is not as high as that of gas lamps.
In recent years, there are electric inoculation ring sterilizers on the market, which are safer to use, and when sterilized, the bacteria on the inoculation ring will not splash, which is more suitable for tuberculosis experimental operations, and the usage of which can be referred to the relevant manuals.
