Osteoarthritis animal model

Summary

Osteoarthritis is a group of clinical diseases caused by various reasons and characterized by the degeneration of articular cartilage as the main pathological feature. Establishing appropriate animal models of osteoarthritis is an important way to study its pathogenesis and prevention methods. Studies have shown that disorders in the biomechanics of weight-bearing joints are one of the main causes of osteoarthritis.

Principle

The rationale behind [animal models of osteoarthritis]:

Osteoarthritis models can take a long time to prepare by the normal wear-and-tear route, but surgical alteration of the mechanics of a joint to create a biomechanical disturbance in the joint may speed up the preparation of osteoarthritis models. The basic principle of selective gluteal muscle cutting osteoarthritis animal model is to cut one of the main muscle groups to maintain the stability of the hip joint, including the complete cutting of the gluteus maximus muscle attached to the iliac crest and the deep gluteus intermedius and minimus muscles, which will lead to the destabilization of the animal's hip joint, and difficulties in standing and walking, and then induce osteoarthritis of the hip joint.

Principle

The basic principle of [Osteoarthritis Animal Model]: It takes a long time to prepare an osteoarthritis model by normal joint wear and tear pathway, but surgical alteration of the mechanics of a certain joint to produce biomechanical disorders of the joint may speed up the preparation of an osteoarthritis model. The basic principle of selective gluteal muscle cutting osteoarthritis model is to cut the gluteal muscle, which is one of the main muscle groups to maintain the stability of the hip joint, including the complete cutting of the gluteus maximus muscle attached to the iliac crest, and the gluteus intermedius and minimus muscles in the deep part, which will cause instability of the animal's hip joint, and difficulties in standing and walking, and then induce osteoarthritis of the hip joint.

Materials and Instruments

Equipment: Male adult guinea pig.
Reagent: 1% sodium pentobarbital (40 mg/kg).

Move

The basic modeling method of osteoarthritis can be divided into the following steps:
A. Adult male guinea pigs are anesthetized intraperitoneally with 1% sodium pentobarbital (40 mg/kg).
B. The gluteus maximus muscle and its deeper gluteus medius and minimus muscles attached to the iliac crest are completely severed under aseptic conditions, with 1 cm of distal severed ends.
C. The animals are allowed to move freely after the operation. After surgery, the animal will show typical lameness, and the paw prints will become lighter and smaller.
D. Histologic examination reveals cartilaginous changes, which may result in the formation of dense bone tissue on the articular surface.
