Overview

  Denatured globin bodies, also known as hemoglobin inclusions, are essentially denatured hemoglobin particles that typically adhere to the cell membrane. They often occur in susceptible individuals after taking certain medications or exposure to chemicals, which can induce hemoglobin denaturation. Such inclusions may also be observed in patients with unstable hemoglobin disorders. Therefore, they hold diagnostic significance for conditions such as glucose-6-phosphate dehydrogenase deficiency and unstable hemoglobinopathies. After staining, these inclusions appear as small blue-black dots under the microscope. This reagent is intended for research purposes only and is not suitable for clinical diagnosis or other applications.

Procedure (for reference only)

  1. Take 0.5 ml of Heinz body staining solution (crystal violet method) and add 2–3 drops of fresh peripheral blood, then mix well.

  2. Incubate in a 37°C water bath for 5 minutes.

  3. Place one drop of the mixture onto a glass slide, cover with a coverslip, and observe under oil immersion.

  4. Counting: Examine 500–1,000 red blood cells, count the number of Heinz body-positive red blood cells, and calculate the Heinz body positivity rate.

Precautions

  1. Normally, Heinz bodies are absent or only a few tiny Heinz bodies may occasionally be observed.

  2. In general, Heinz bodies are located on the red blood cell membrane or scattered within the red blood cells.

  3. When using this reagent for the first time, it is recommended to perform a preliminary test with 1–2 samples.

  4. For your safety and health, please wear a lab coat and disposable gloves during operation.

  5. After opening, the reagent should be used as soon as possible to avoid affecting subsequent experimental results.
