Lactobacillus Agar Medium preparation method: Weigh 45.5 g of this product in 1 L of distilled or deionized water (the volume may be scaled up or down proportionally), dissolve with gentle warming, and dispense; autoclave at 121°C for 15 minutes; after sterilization, shake well to prevent agar from settling and solidifying at the bottom of the vessel, then set aside for use. Lactobacillus Agar Medium formula (per liter) contains: Peptonized milk 15.0 g (provides nitrogen source and growth factors), yeast extract powder 5.0 g (supplements B vitamins and amino acids), tomato extract powder 2.5 g (provides specific growth-promoting factors), glucose 10.0 g (serves as the primary carbon and energy source), potassium dihydrogen phosphate 2.0 g (maintains pH buffering system), Tween 80 1.0 g (acts as an emulsifier and fatty acid source to promote lactobacillus growth), and agar 10.0 g (serves as a solidifying agent); final pH adjusted to 6.8 ± 0.2 (25°C).
