Overview

Mitochondrial respiratory chain complex V, also known as F₁F₀-ATP synthase, consists of two subunits, F₁ and F₀. It is a key enzyme for ATP synthesis in mitochondrial oxidative phosphorylation and chloroplast photophosphorylation, widely present in the mitochondria of animals, plants, microorganisms, and cultured cells. This enzyme utilizes the proton electrochemical gradient generated by the respiratory chain to catalyze ATP synthesis and can also hydrolyze ATP through a reverse reaction. The detection principle of this kit is based on the hydrolysis of ATP by mitochondrial respiratory chain complex V, producing ADP and inorganic phosphate (Pi). In the presence of a reducing agent, Pi reacts with ammonium molybdate to form a dark blue phosphomolybdenum complex. By measuring the change in absorbance at 660 nm, the activity of mitochondrial respiratory chain complex V in the sample can be calculated.
