Xylanase (EC 3.2.1.8) is mainly produced by microorganisms and catalyzes the hydrolysis of xylan. It is also known as pentosanase or hemicellulase. This enzyme can degrade cell wall components and β-glucans in raw materials used in the brewing and feed industries, reducing viscosity during the brewing process and promoting the release of active substances. At the same time, it reduces non-starch polysaccharides in feed, facilitating nutrient absorption and utilization. Therefore, it is widely applied in brewing and feed industries. Neutral xylanase (NEX) is generally isolated from microorganisms with an optimal growth pH in the range of 6–8. The assay principle of this kit is as follows: NEX catalyzes the degradation of xylan into reducing oligosaccharides and monosaccharides under neutral conditions. The reducing sugars then react with 3,5-dinitrosalicylic acid (DNS) in a boiling water bath to generate a colored product with a characteristic absorption peak at 540 nm. The color intensity of the reaction solution is proportional to the amount of reducing sugars produced by the enzymatic hydrolysis. By measuring the rate of increase in absorbance at 540 nm, the enzyme activity of NEX can be calculated.
