Overview
Peroxy Orange 1 (PO1) is a novel hydrogen peroxide (H₂O₂)-specific probe, which can be combined with APF, a green fluorescent highly reactive oxygen species (hROS) probe. Peroxy Orange 1 also serves as a live-cell dye.As a highly sensitive fluorescent probe for H₂O₂, Peroxy Orange 1 emits orange fluorescence with an excitation wavelength of 543 nm and an emission wavelength of 565 nm.Peroxy Orange 1 contains boronate ester groups, which can be oxidized into phenol groups by H₂O₂ to produce bright orange fluorescence.This boronate-phenol transformation mechanism does not react with other reactive oxygen species (ROS) such as superoxide anions and NO, thus guaranteeing the high selectivity of Peroxy Orange 1 toward H₂O₂.Peroxy Orange 1 exhibits excellent cell permeability, enabling the detection of intracellular H₂O₂ in live cells with negligible cytotoxicity.It is compatible with conventional green fluorescent probes including GFP and APF, and suitable for multicolor fluorescence imaging assays.
