Overview

  Negative staining, also known as negative contrast staining, is a staining technique discovered by Hall as opposed to conventional staining (i.e., positive staining). The principle involves surrounding low-electron-density samples with heavy metal salts to increase the electron density around the sample, thereby creating mass-thickness differences among fine structures and enhancing the scattering-absorption contrast. This allows the sample to appear as bright structures against a dark background. Common negative staining agents include phosphotungstic acid, ammonium molybdate, and India ink, among which 1–3% phosphotungstic acid is the most frequently used.

  Phosphotungstic Acid Ethanol Solution (3%) is primarily composed of phosphotungstic acid and ethanol. Phosphotungstic acid serves as the active component for staining sperm maturity and sperm nuclear histones. This reagent is intended for research purposes only and is not suitable for clinical diagnosis or other applications.
