STATs are cytosolic proteins with a common structure consisting of an N-terminal oligomerization domain, which favors formation of STAT dimers, followed by a DNA-binding domain and a C-terminal SRC homology-2 (SH2) domain, which is involved in association between STATs and receptors (PMID:22383755). The DNA-binding domain can bind to DNA which affects its migration in Western blot experiments.

IF analysis of Recombinant STAT4 Antibody (Ab129436) in HeLa cells. The cells were fixed and permeabilized with 100% methanol for 5 minutes, and blocked with 5% BSA for 1 hour at room temperature. Cells were stained with Recombinant STAT4 Antibody (Ab129436) at 1/200 dilution in blocking buffer overnight at 4°C, and then incubated with Goat Anti-Rabbit IgG H&L (AF488) (Ab181954) at a dilution of 1/1000 for 1 hour at room temperature in the dark (green). Cells were counterstained with DAPI (blue).
