Toluidine blue staining test

Summary

Toluidine blue is one of the commonly used synthetic dyes, belonging to the quinone imine dyes, which generally contain two chromophores, an amine group and a quinone-type benzene ring, to constitute the chromophore to develop color. In addition to the chromophore, the dye must also have an atomic group that enables the chromophore to have an affinity for tissues and other dyed materials, i.e., the auxiliary chromophore.

1. Dilute the Toluidine Blue Staining Solution with water (starting with a 1:100 dilution according to the desired degree of staining, to be determined by experimental experience).2. Soak the slide briefly in diluted staining solution, then soak it several times in water and press-fix it according to the selected method.
