Protease phosphatase inhibitor mixture (generic, 50X) (Protease and phosphatase inhibitor cocktail for general use, 50X) is a general-purpose protease and phosphatase inhibitor cocktail for cell or tissue protein extraction. This product contains one bottle of protease inhibitor mixture (50X), one bottle of phosphatase inhibitor mixture (50X), and one bottle of 0.05M EDTA.
Samples such as cell or tissue extracts contain many endogenous proteases and phosphatases, which can easily lead to protein degradation or demodification in the extracts, thus affecting subsequent protein detection. Therefore, adding appropriate protease, phosphatase and other inhibitors in extracts and other samples is an effective way to prevent protein degradation and demodification.
This product is an optimized and tested mixture of protease and phosphatase inhibitors for routine cell or tissue protein extraction, including a broad spectrum of serine, cysteine and acid protease inhibitors/aminopeptidase inhibitors, as well as serine/threonine, tyrosine, acid and alkaline phosphatase inhibitors.
One package of this product is usually sufficient for the preparation of 500ml of lysate.
Operation process:
The protease phosphatase inhibitor mixture is a 50X storage solution, which is added to the lysate in a ratio of 1:50 (e.g., 20μl protease inhibitor mixture and 20μl phosphatase inhibitor mixture in 1ml lysate), mixed and ready for use. The lysate containing protease phosphatase inhibitor mixture should be used on the spot, and should not be frozen after preparation for subsequent use. As needed, 0.05M EDTA is also added to the lysate in a ratio of 1:50. After the required inhibitors have been added and blended, routine cell or tissue lysis and protein extraction can begin.
Note:
1. This product can be stored after proper packaging, and should be added to the appropriate solution such as cracking solution before use.
2. this product provides independent packaging of EDTA, can be used selectively. For example, it is subsequently used to detect metalloproteinase activity in the extract, otherwise EDTA should not be added.
3. the protease and phosphatase inhibitors contained in this product are harmful to human body, please be careful when operating, do an effective protection.
