Descripción general

There are various methods for neutral fat staining in the laboratory, such as Sudan Black B, Sudan Ⅱ, Sudan Ⅲ, Sudan Ⅳ, and Oil Red O staining. Sudan dyes are generally used in traditional methods, while the azo dye Oil Red O is now more suitable for fat staining. Oil Red O is a highly effective lipid solvent and lipochrome; although its binding affinity to phospholipids is relatively weak, it readily binds to triglycerides to form small lipid droplets. Sudan Black B Stain is mainly used to demonstrate abnormal deposition of lipids, fatty degeneration of tissues and organs, as well as the fatty degeneration commonly occurring in parenchymal organs such as the liver, kidneys, and heart, where numerous neutral fat droplets appear within cells. It also aids in the identification and diagnosis of tumors arising in adipose tissue and their pathological properties. During the experiment, fixatives containing ethanol should not be used for specimens. If fixation is necessary, 10% formalin can be applied. In addition, paraffin sections are not recommended; instead, carbowax sections or frozen sections should be adopted.

Operating Procedures (For Reference Only)

1. repare cryosections of fresh tissues at a temperature typically ranging from -20℃ to -25℃. For lipoma specimens, adjust the temperature to -30℃.

2. Cut frozen sections with a thickness of 5–10 μm (6–8 μm is optimal) and mount them on glass slides.

3. Rinse the sections briefly with 70% ethanol.

4. Immerse the sections in Sudan Black B Stain for staining.

5. Wash away excess stain with 70% ethanol.

6. Rinse the sections with distilled water for 1 minute.

7. Stain the cell nuclei lightly with Nuclear Fast Red Stain.

8. (Optional) If overstaining is observed under the microscope, differentiate the sections with acid differentiation solution for a few seconds.

9. Rinse briefly with tap water, followed by a quick rinse with distilled water.

10. Blot excess water from the sections and their surroundings with filter paper and allow them to air-dry slightly.

11. Mount the sections with glycerin jelly or gum arabic.

Phospholipids Grayish: Black

Cell Nuclei: Red


Precautions

1. Do not use ethanol-containing fixatives or prepare paraffin sections for specimens; frozen sections are required.

2. To prevent dye precipitation during staining, seal the container when immersing sections in the stain. Avoid contact with flowing air to prevent precipitation caused by solvent evaporation.

3. Frozen sections stain easily. Avoid overstaining when counterstaining with Mayer's hematoxylin.

4. Sudan dyes are prone to fading and should be stored in a sealed environment.

5. Specimens mounted with glycerin jelly have a short shelf life. For long-term preservation, seal the edges between the coverslip and slide with neutral balsam.
