Descripción general

Lipids are a general term for neutral fats, lipids, and their derivatives. Their common physical characteristic is insolubility in water and solubility in organic solvents (e.g., ethanol, ether, etc.). There are two main types of fats in the human body: Storage fats: such as neutral fats, mainly distributed in subcutaneous tissues, kidneys, pancreas, and other parts. Structural fats: such as lipids (phospholipids, glycolipids, cholesterol, etc.), mainly distributed in cells. Neutral fats are lipids composed of three molecules of fatty acids and one molecule of glycerol, and they are neutral in nature. Staining methods for neutral fats often include Sudan Ⅱ, Sudan Ⅲ, Sudan Ⅳ, Sudan Black B, and Oil Red O staining. Traditional methods use Sudan dyes, but recent studies have found that the azo dye Oil Red O is more suitable for fat staining. Oil Red O is a strong lipid solvent and lipochrome; it binds easily to triglycerides to form small lipid droplets, but has a relatively weak binding affinity for phospholipids.

Sudan Black B Staining Solution (Cell-specific), abbreviated as SBB Stain, works on the principle that Sudan Black B is a liposoluble dye. It can dissolve in lipid-containing structures in the cytoplasm, staining fats, phospholipids, cholesterol, and other lipids brown-black or dark black and localizing them in the cytoplasm. Sphingomyelin, cephalin, and other substances show positive reactions after SBB staining; conversely, clear cell renal cell carcinoma, ovarian fibroma, and other tissues show negative reactions after SBB staining.

Preparation before use: Mix Reagent B and Reagent C to prepare SBB Stain. The stain should be used immediately after preparation.

Materials to be prepared by the user:

1. Glass slides

2. 70% ethanol

3. Distilled water

4. Microscope


Operating Procedures (For Reference Only)

1. Prepare smears of fresh bone marrow or blood, and fix them with SBB Fixative.

2. Take out the smears and air-dry them in circulating air.

3. Immerse the smears in freshly prepared SBB Stain and incubate at room temperature.

4. Transfer the smears to Sudan Black B Stain for further staining.

5. Wash away excess stain with ethanol, followed by rinsing with distilled water.

6. Perform counterstaining with Wright Stain.


Staining Results

Neutral Fats: Brown-black or dark black granules

Cell Nuclei: Red

In the granulocyte series, except for early myeloblasts (negative), cells from the well-differentiated myeloblast stage onwards show progressively stronger positive reactions with cell maturation. Senescent neutrophils show weakened positive reactions. Monocytes show weak positive reactions. Lymphocytes show negative reactions. Plasma cells and megakaryocytes are both negative. Eosinophils and Auer bodies show strong positive reactions.

Negative Control: Treat the same sample with a mixture of equal volumes of ether, chloroform, and acetone to dissolve general fats; then use pre-cooled acetone to dissolve phospholipids. Subsequent SBB staining of the treated sample should yield negative results.


Precautions

1. Do not use ethanol-containing fixatives for specimen preservation.

2. Sudan dyes are prone to fading and should be stored in sealed containers.

3. To prevent dye precipitation, place the smears in a petri dish with small bamboo sticks at both ends, and stain the smears with the blood film facing downward (suspended above the stain solution).

4. Sphingomyelin and cephalin yield positive results with SBB staining, which is helpful for the diagnosis of lipid storage diseases.

5. For your safety and health, wear a lab coat and disposable gloves during operation.
